Ethidium Bromide Solution can be used to detect both single and double stranded nucleic acids (both DNA and RNA). However, Ethidium bromide ( EtBr ) does not intercollate into single-stranded DNA very well. Therefore, the fluorescence of single-stranded DNA is relatively poor in comparison to double-stranded DNA.

Use 2.5 µl of ethidium bromide solution for every 50 ml of molten agarose, running buffer, or staining solution which is about one drop.

The recommended final concentration of ethidium bromide in an agarose gel and/or electrophoresis buffer is 0.5 µg/ml.

Recommended Use

A commonly accepted volume for one drop of liquid is 0.05 ml or 50 µl. (20 drops = 1 ml)

How do I make a stock solution with a concentration 0.5 µg/ml?

Add one drop (approximately 50 µl) of IBI 10 mg/ml Ethidium Bromide Solution to 1-liter of DDI water to make a stock solution with a desired concentration of 0.5 µg/ml. This stock solution of ethidium bromide can then be added to the researcher’s agarose or buffer solution.

How much of the stock solution should be used with an agarose or buffer solution?

For a 50 ml agarose or buffer solution, add 2.5 µl of the stock ethidium bromide solution (0.5 µg/ml).
