The gMAX Genomic DNA Mini Kit has been optimized for genomic, mitochondrial, and viral DNA purification from whole blood (fresh and frozen) tissue, formalin-fixed paraffin-embedded tissue, cultured cells, buccal swab, amniotic fluid, hair, and insects in one convenient kit. This DNA extraction kit uses Proteinase K and chaotropic salt to lyse cells and degrade protein, allowing DNA to bind to the glass fiber matrix of the spin column. Contaminants are removed using a wash buffer and the purified genomic DNA is eluted by a low salt elution buffer, TE, or water. The entire procedure can be completed in less than 30 minutes without phenol/chloroform extraction or alcohol precipitation. The purified DNA is suitable for use in PCR or other enzymatic reactions.

The quality of the gMAX Genomic DNA Mini Kit is tested on a lot-to-lot basis by isolating genomic DNA from 200 µl of whole human blood. The purified DNA (5 µg with an A260/A280 ratio of 1.8-2.0) is quantified with a spectrophotometer and analyzed by electrophoresis.

Can QIAGEN reagents be used with IBI columns?Yes, anything that is “lysis-based” will work with IBI columns.

Can I extract Genomic DNA from a Milk sample using this kit?Yes this kit will work when using a Milk sample. For small samples (200 µl) use the blood protocol. For larger samples (1 ml) use the amniotic fluid protocol.

When the tissue sample is slightly degraded, what can I do to improve the low yield of gDNA?1. After adding the GT buffer and Pro K to the sample, proceed to vortex, and then incubate over night at 60°C. During the overnight incubation use an automated shaking water bath to continually agitate the sample during incubation. 2. Perform the RNA removal step as recommended in the protocol. 3. Pre-heat the elution buffer to 60°C for the elution step. 4. Using large amounts of elution buffer (100 µl – 200 µl) during the elution step can raise the DNA yield. 5. Homogenize tissue samples prior to lysis to help in the Pro-K digestion.

What is the typical tissue yields using the gMax kits?Depending on the starting material, it can be anywhere from upwards of 50µg with 25 mg of tissue, or between 10-40µg with 20 mg of tissue.

Where can I get more of the Proteinase K Solution? You can purchase our Proteinase K Solution-5 ml (IB05406 20 mg/ml) and dilute it with purified water (ddH20) to a concentration of 10 mg/ml.
