Ponceau S protein stain is used to visualize proteins on a nitrocellulose or PVDF membrane. To create a Ponceau S solution, dissolve 0.5g of Ponceau S in 1ml of glacial acetic acid and bring to 100ml with ddi water.

To stain with Ponceau S, remove blot and rinse with ddi water. Then, stain with Ponceau S solution for 1 – 2 minutes. Rinse for 3 – 5 minutes with ddi water. Red bands should appear against the white background.
To reversal stain with Ponceau S solution, place the nitrocellulose membrane in Ponceau S solution for 5 minutes at room temperature. Destain in ddi water for 2 minutes and mark the molecular weight standard with indelible ink. To complete the destain, soak in water for 10 minutes. Alternatively, use 0.1M NaOH solution for 5 minutes if proteins of interest are not basic.
