IBI provides the phenol chloroform solution at a pH of 6.7 or 8.0 ± 0.2 for use in typical DNA purification.

To use, add an equal volume of Phenol│Chloroform Isoamyl Alcohol to the digested DNA solution to be purified in a 1.5 ml micro-centrifuge tube. Mix gently for five minutes. Centrifuge at 10,000 rpm for three minutes. The phases should be well separated. Carefully remove the aqueous layer containing DNA and place into a new tube. Repeat prior steps until interface is no longer visible.

Recommended Use
IBI provides Phenol-Chloroform Solution at a pH of 6.5 – 6.9 for use in typical DNA purification. Example: To purify 400 μL of sample, add 400 μL of Phenol-Chloroform solution.

IBI’s Phenol-Chloroform Solution is packaged at a standard pH of approximately 6.7, but can be increased to approximately 8.0 when mixed with the supplied 26 mL bottle of Tris. Pour the entire bottle of buffer into the bottle of phenol-chloroform.

Storage Store at +4°C. Warm to room temperature before opening. Keep tightly sealed. Phenol Chloroform Solution is prepared from ultra-pure, double-distilled phenol, chloroform, and isoamyl alcohol at a ratio of 25:24:1 respectively. The product is packaged under an inert gas for maximum stability.

Is there something wrong with the Phenol-Chloroform solution if the upper layer gets milky? No, the Phenol-Chloroform solution is still good to use.
