Ethidium bromide can be used to detect both single and double stranded nucleic acids (both DNA and RNA). However, Ethidium bromide does not intercollate into single-stranded DNA very well. Therefore, the fluorescence of single-stranded DNA is relatively poor in comparison to double-stranded DNA.

The recommended final concentration of ethidium bromide in an agarose gel and/or electrophoresis buffer is 0.5 µg/ml.
