DNA Isolation from Yeast

The gYEAST Genomic DNA Reagent Kit has been optimized to provide superior yield and purity. The kit offers a simple and gentle reagent DNA precipitation method for isolating high molecular weight genomic, mitochondrial, or viral DNA from Saccharomyces cerevisiae and a variety of other fungus species. This highly versatile solution based system can be scaled proportionately in order to satisfy larger yeast sample sizes, providing a convenient procedure with minimal hands-on time. The provided sorbitol buffer, when combined with β-mercaptoethanol, zymolase, or lyticase will efficiently lyse fungus cell walls consisting of chitin or polysaccharides. The extracted DNA is suitable for use in PCR or other enzymatic reactions.

The gYEAST Genomic DNA Kit is tested on a lot-to-lot basis. Saccharomyces cerevisiae (2 x 108) is harvested by centrifugation at 5,000xg for 10 minutes. A 15µl aliquot of purified genomic DNA from a 100µl eluate is analyzed by electrophoresis on a 1% agarose gel.
